Old setup, need a new Graphics Card for Diablo 3

you should be fine with getting an hd 6570.

dont get something like hd 7750 your cpu will bottlenck it go for hd 6570its good for both cpu and psu.

Also, the mobo has a PCI Express x16 slot, that means I should be okay with PCIe 2.0, 2.1 etc?

yes you will be okay with 2.1 pcie.
