5 second long beep sound then 2 second rest from cpu,what may be the defect

Video or RAM, try re-seating both and cold booting.

Sounds like RAM to me. Try another slot and put your ram in there, sometimes certain slots are faulty. If the system boots its either your motherboard having issues or you didn't seat the RAM correctly in the right slot.
