I am going to build a PC for my dad who is a graphic designer and was wondering what graphics card is best for the job, i know it is low usage work but would a Nvidia quadro card be good for it or is a GT 640 or something better to use?

Ofcourse Quadro.
GTX or GT series aren't for Designing. They are for Gaming.

Photo editing software is evolving to utilize OpenCL, OpenGL and Direct Compute on graphics cards. Additionally, nVidia offers CUDA processing on certain cards.

In editing, CUDA tends to focus primarily on color correction of images.

OpenCL/GL acceleration tends to benefit specific functions by vendor. The best all-around OpenCL/GL card for GPU compute would be the FirePro V4900.

I honestly don't know where MS is going with Direct Compute.

What kind of graphic design are we talking about? Photoshop and other 2D / 3D modelling for entertainment like 3Ds Max / 3D design like AutoCad

Mostly photoshop, the main thing needed is accurate colour rendering.

I'm no expert in that, but I think the main concern should be the display itself rather than the gpu. One think to keep in mind is that the card should have appropriate connections
