I'd like to disconnect my wireless base station from my
desktop (which is on the top floor of my home) and connect
the router to an incoming cable on the middle floor of my
home so my wife's laptop in the basement gets a better
signal. If I connect a wireless USB adapter to my
desktop, will my desktop PC be able to access the internet?

Wi-Fi signals are radio signals and is difficult to
predict how far they cross walls or deal with
interference (e.g. refrigerator, microwave...)
Borrow your wife's laptop and check if it can get
wireless access from the place where you are planning to
place the base station, or better...
- Place the base station there and use your wifes laptop
to see if the signal reaches the location of the
desktop), and
- that the signal also reaches the basement
