I've used to connect external audio to mobo's line input and to listen to music or whatever, while being able to capture (record) the input signal instantly. So it used to be with all mobos I have used until now.
Now it appears that new Intel DQ35JO mobo with Realtek chipset doesn't support direct link from line in to line or headphones output.
I was rather suprised, when Intel representative told me that it's by design and possible only with help of 3rd party software.
For excample Avermedia USB TV box with audio output can't be connected to line in, but audio from USB box must go directly to headphones and for recording the cables must be swapped.
