"Best" game is hard to judge as there are lots of games out there that are
good but they simulate much different aspects of space combat. Its an
apples to oranges kinda thing.

I'll ask you a few questions in return to find out what kind of game you
want to play and then we can find the game that best plays your interests.

Big ships or fighters?
A few ships or a fleet of ships?
Lots of control over the ship's systems or minimal control?
Cinematic movement (Star Trek) or realistic vector movement (Babylon 5)?
Construction rules or pre-made universe?
Hex board or open table?
Destroy ships or capture ships?
2 hour battles or 6 hour battles?
Famous TV or movie setting or generic game?
Popular game or small following?
Etc, etc.

You can find out more about all the space combat games currently available
at my website (URL below)

OK, per your answers, it sounds like Silent Death is the game you are
looking for.
http://store.ironcrown.com/category.jsp?category=2043
Silent Death is currently a bit beyond its prime. It has had lots of
supplements released but due to the bankruptcy of ICE among other things,
nothing new has been released in a while. ICE has plans to continue
supporting Silent Death but many fans are still waiting.

As a result, some of the former developers have created a new game,
Arclight.
http://secondrat.com/arclight.php
Arclight draws from Silent Death and many other games and plans to allow
gamers to help develop the universe. It just released in July so it is
still very new to the gaming scene.
