As long as your boot order isn't set to boot from a USB device, they can be plugged in at either time.

Most of USB devices work in plug & play system, which means that in the moment you connect them to the computer you can immediately start working with them.

What can be deduced from it, is that unless the system asks you to restart after installing a USB device, it doesn't matter if the computer is turned on or not when connecting the USB.
