I have the Router so that my second computer can also connect with my Comcast
Cable. I have the modem connected directly to this laptop, and wish to
connect my second computer through the router. Currently I have my modem
connected through the USB link provided by the modem into my laptop. Can I
still connect my modem into the Router through the ethernet cables and then
have the second computer connected wirelessly or do they both need to go
through the router?

If you have a wireless adapter (like built into the laptop), all you need do
is create the connection in Network Connections. The only time you need
directly connect via RJ-45 Ethernet cable is if you don't have a wireless
adapter.
