Error message 16 bit Windows Subsystem

c:\WINDOWS\SYSTEM32\AUTOEXEC.NT.The system file is not suitable for running
MSDOS and Microsoft Windows applications.

Also:Error Message: Entry Point Not Found. (running Works)

These two error messages have suddenly started to appear.I have re-installed
XP as i have a couple of applications that are DOS based that I use
regularly.Reinstalling solved the problem for a while then problem treturned.
Any assistance would be gratefully accepted.
