Currently have an i5 3570k on my MSI z77a-gd65 motherboard, but I'm downsizing to an mITX Asus P8Z77-i Deluxe. I was wondering if I downloaded the new Asus drivers while still using the MSI board, then disconnected everything and re-installed all hardware on the Asus board (including same ssd w/ Windows 7), will everything work fine? Or do I need to buy a new key?

Tl;DR do I need get a new Windows 7 key if I only change motherboard?

You will use the same key, if doesn't work open Windows activation troubleshooting it will give you some long numbers you just call Microsoft and ask him to activate Windows Key on this PC just he will ask you to tell him the numbers and he will give you other numbers type it in and done. Did it before just if it's work choose it for the best solution :)
Best regards

Have you got windows on your current drive and will you be using that exact drive when you get your new motherboard? If so then you don't need to call Microsoft. The OS is on the drive not on the motherboard.
