Best way to hook monitor to graphics card?

DVI to DVI. HDMI has sound, rarely anyone needs that and DisplayPort tend to cost more, DVI is your cheapest and best choice.

DVI and DisplayPort offer 144Hz refresh rates and little latency (won't even notice). Same Video quality as HDMI
HDMI offers 60hz max refresh rates and little latency (won't even notice)

So if I need to use the speakers in the monitor, I should use HDMI?
 
yes, or a 3.5mm to 3.5mm audio jack if you use DVI, that is if the monitor has an audio input also.

Be aware;

Some monitors don't allow HDMI to use as their Audio source, some have separate audio connections to use instead, and the Audio form HDMI would be utterly useless.

If you see a 3.5mm audio input, your monitor may not use HDMI as the sound input.
