Best BIOS update methods to ensure success?

Old mobo: Z77A-G45 Thunderbolt

I updated the BIOS after three years of never updating it. Skipped a couple BIOS revisions, assuming this is a fatal error. See post:

Since flashing that BIOS I have done so many different troubleshoots that I have to assume the old mobo is beyond help. They include:
1. clearing CMOS
2. replacing 2032 mobo battery
3. removing a stick of RAM
4. unplugging hdds and dvd drive and plugging in a flash drive (FAT32) with BIOS on the root of the flash drive, then trying to power up the mobo so that it reads the flash drive's BIOS file.

5. connected a different power supply

I plan on getting an Asus X99 A-II. What are people's preferences on updating the BIOS? What steps are needed to make absolutely sure nothing goes wrong? Any tips or feedback would be awesome! Just trying to learn as much as I can.

**I've looked at the manual and there are a couple of options updating the BIOS for this Asus mobo. Does anyone have a preference for this particular model?
